CD45R, also designated CD45 and PTPRC, is identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. The melanoma-associated antigen (MAGE) family consists of a number of antigens recognized by cytotoxic T lymphocytes. The MAGE genes were initially isolated from different kinds of tumors, and based on their virtually exclusive tumor-specific expression in adult tissues, they have been used as targets for cancer immunotherapy. MAGE genes encode for tumor-rejection antigens and are expressed in tumors of different histologic types, but not in normal tissues, with the exception of testis and placenta. Anti-cytokeratin clone PCK/3150 demonstrates a broad spectrum of cytokeratin reactivity. In normal tissues, PCK/3150 is reactive with most epithelial types, including bile ducts and hepatocytes in liver, bladder epithelium, breast ducts, bronchial epithelium, endometrium, intestinal epithelium of stomach, duodenum, ileum, colon, rectum, pancreas, ovarian epithelium, pancreatic acini, pituitary acini, pneumocytes, prostate, thyroid, skin (positive on the basal layer and negative on the superficial layers of squamous epithelium), and apocrine and sweat glands. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Beta-2 microglobulin is a component of the class I major histocompatibility complex (MHC I) where it functions in antigen processing and presentation. Involved in the presentation of peptide antigens to the immune system. Human Beta-2 microglobulin is expressed on many types of cells including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial cells, and epithelial cells. It is absent on erythrocytes. This MAb is specific to human Beta-2 microglobulin and does not react with non-human primate cells. This antibody reacts with all cell types excluding erythrocytes. Bcl-6 is a transcriptional repressor mainly required for germinal center (GC) formation and antibody affinity maturation which has different mechanisms of action specific to the lineage and biological functions. This antibody is a mixture of four different monoclonal antibodies. This antibody is specific to 60 kDa latent membrane protein (LMP-1) encoded by the BNLF1 gene of the EBV. Each clone reacts with different epitopes on the hydrophilic C-terminus of the cytoplasmic domain of LMP-1. This antibody stains strongly with EBV-positive lymphoblastoid cell lines and EBV infected B cell immunoblasts in infectious mononucleosis. EBV, also designated human herpesvirus 4 (HHV-4), is a member of the herpesvirus family and is one of the most common human viruses. EBV infects B cells and, though often asymptomatic, it can cause infectious mononucleosis, a disease characterized by fatigue, fever, sore throat and muscle soreness. This antibody recognizes a protein of 39 kDa, which is identified as Aurora kinase B (or Aurora B). Aurora B is a serine/threonine protein kinase and component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. It is important for accurate chromosome segregation, cytokinesis, protein localization to the centromere and kinetochore, correct microtubule-kinetochore attachment, and regulation of the mitotic checkpoint. Aurora B forms a tight complex with inner centrosome protein and survivin. Inactivation of any of these proteins causes similar defects in chromosome segregation. A significant overexpression of Aurora B has been found in a variety of human tumors including non-small cell lung carcinoma, astrocytoma, seminoma and carcinomas of the colon, prostate, endometrium and thyroid. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Cytokeratin 15 is a specific marker of stem cells of the hair-follicle bulge and may be a useful marker for diagnosis between basal cell carcinoma (BCC) and trichoepithelioma. Trichoblastoma are benign neoplasms of follicular differentiation frequently found in nevus sebaceous. Many morphologic features are shared with nodular basal cell carcinoma, sometimes rendering a diagnosis difficult. BarX1 is a member of the Bar subclass of homeobox transcription factors. Studies of the Mouse and chick homolog suggest the encoded protein may play a role in developing teeth and craniofacial mesenchyme of neural crest origin. The protein may also be associated with differentiation of stomach epithelia. Down-regulation of Barx1 promotes hepatocellular carcinoma cell (HCC) migration, invasion and metastasis, whereas up-regulation of Barx1 inhibits HCC migration, invasion and metastasis.
